Pharmacological action

A drug that activates the metabolism in tissues, reduces tissue hypoxia, improves trophism and stimulates the regeneration process. It is a hemoderivate that is obtained by dialysis and ultrafiltration (compounds with a molecular weight of less than 5000 daltons penetrate).

It has a positive effect on the transport and utilization of glucose, stimulates oxygen consumption (which leads to stabilization of the plasma membranes of cells during ischemia and reduces the formation of lactates), thus having an antihypoxic effect.

Actovegin not only increases the intracellular glucose content, but also improves oxidative metabolism, thereby improving the energy supply of the cell, as evidenced by an increase in the concentration of direct carriers of free energy, such as ATP, ADP, phosphocreatine, as well as amino acids, such as glutamate, aspartate and GABA.

In cases of metabolic disorders and blood supply to the brain, such as cerebral insufficiency syndrome (dementia), glucose transport through the BBB and its utilization by cells deteriorates. PDH activity and acetylcholine concentration are also reduced. The use of Actovegin contributes to the normalization of these indicators, improves the transport and utilization of glucose, while increasing oxygen consumption is observed.

It has been shown that Actovegin also acts in the same way in peripheral (arterial, venous) circulatory disorders and the corresponding consequences of such disorders (arterial angiopathy, ulcerative lesions of the lower extremities), as well as when used to accelerate wound healing. In ulcers of various etiologies, trophic disorders (bedsores), burns and radiation damage, Actovegin improves not only morphological, but also biochemical parameters of granulation, for example, increases the concentration of DNA, hemoglobin and hydroxyproline.

The effect of Actovegin begins to appear no later than 30 minutes after ingestion and reaches a maximum on average after 3 hours (2-6 hours).

Indications

• in the complex therapy of metabolic and vascular disorders of the brain (various forms of cerebral circulatory insufficiency, dementia, traumatic brain injury);
• peripheral (arterial and venous) vascular disorders and their consequences (angiopathy, trophic ulcers).

Contraindications

• hypersensitivity to the drug.

With caution, the drug should be prescribed for stage II-III heart failure, pulmonary edema, oliguria, anuria, hyperhydration, diabetes mellitus, hyperglycemia.

Side effects

Allergic reactions:  urticaria, edema, drug fever.

Interaction

Drug interaction of Actovegin has not been established.

How to take, course of use and dosage

Assign 1-2 tablets 3 times/day before meals.

The tablet is not chewed, washed down with a small amount of water.

The duration of treatment is 4-6 weeks.

Overdose

Information about overdose of Actovegin was not provided.

Special instructions

If allergic reactions develop, Actovegin should be discontinued. If necessary, standard therapy for allergic reactions (antihistamines and/or corticosteroids) is performed.
