Pharmacological action
A broad-spectrum antimicrobial agent belonging to the nitrofuran group. Resistance to furazidine develops slowly and does not reach a high degree. Active against gram-positive and gram-negative microbes, pathogenic strains Staphylococcus spp. and other antibiotic-resistant microorganisms.
It is effective against gram-positive cocci (streptococci, staphylococci), gram-negative bacilli (Eschirihia coli, Salmonella, shigella, Proteus, klebsiella, Enterobacteria), protozoa (Giardia). Furazidine is more active against staphylococci, Escherichia coli, Aerobacter aerogenes, Bacterium citrovorum, Proteus mirabilis, Proteus morganii, compared to other nitrufurans. Furazidine is more active against Enterococcus faecalis, Staphylococcus spp. compared to other groups of antimicrobials.
Against most bacteria, the bacteriostatic concentration ranges from 1: 100,000 to 1: 200,000. The bactericidal concentration is approximately twice as high. Under the influence of nitrofurans in microorganisms, the respiratory chain and the tricarboxylic acid cycle (Krebs cycle) are suppressed, as well as other biochemical processes of microorganisms are inhibited, resulting in the destruction of their shell or cytoplasmic membrane.
As a result of the action of nitrofurans, microorganisms secrete fewer toxins, and therefore an improvement in the general condition is possible even before the pronounced suppression of microflora growth. Nitrofurans, unlike many other antimicrobial agents, not only do not depress the body’s immune system, but on the contrary, activate it (increase the complement titer and the ability of white blood cells to phagocytize microorganisms). In therapeutic doses, nitrofurans stimulate leukopoiesis.
Indications
Urogenital infections (acute cystitis, urethritis, pyelonephritis); gynecological infections; skin and soft tissue infections; severe infected burns; for preventive purposes during urological operations (including cystoscopy, catheterization).
Contraindications
Severe chronic renal failure; pregnancy; lactation (breastfeeding); children under 3 years of age; hypersensitivity to furazidine and other nitrofurans.
Side effects
From the digestive system: Â rarely – nausea, vomiting, loss of appetite, liver function disorders.
From the central nervous system and peripheral nervous system: Â rarely-headache, dizziness, polyneuritis.
Allergic reactions: Â rarely – skin rash (including papular rashes).
Interaction
When furazidine is combined with ristomycin, chloramphenicol, and sulfonamides, the risk of hematopoiesis suppression increases (do not use it simultaneously).
It is not recommended to use simultaneously with nitrofurans drugs that can acidify the urine (including ascorbic acid, calcium chloride).
How to take, course of use and dosage
Inside, after eating, with plenty of fluids.
For urogenital infections (acute cystitis, urethritis, pyelonephritis), gynecological infections:
Adults are prescribed 50-100 mg 3 times a day.
Children are prescribed 25-50 mg 3 times a day (but not more than 5 mg/kg of body weight per day).
The course of treatment is from 7-10 days. If you need to repeat the course of treatment, you should take a break for 10-15 days.
For the prevention of infection during urological operations, cystoscopy, catheterization:
Adults are prescribed 50 mg once 30 minutes before the procedure.
Children are prescribed 25 mg once 30 minutes before the procedure.
If you forget to take the next dose, take the next dose at the usual time. Do not take a double dose to replace a missed dose.
Overdose
Symptoms: neurotoxic symptoms, ataxia, tremor.
Treatment: Â in case of overdose, you should drink a large amount of liquid. Antihistamines are used to relieve acute symptoms. For the prevention of neuritis, vitamins (thiamine bromide) can be prescribed.
Special instructions
To reduce the likelihood of side effects, the drug is washed down with a large amount of liquid.
If side effects occur, the drug should be discontinued (toxic effects are more common in patients with reduced excretory function of the kidneys).
If any of the side effects listed in the instructions get worse, or any other side effects that are not listed in the instructions are noted, you should immediately inform your doctor.
Given the possible side effects of furazidine from the nervous system, care should be taken when driving vehicles, as well as when engaging in other potentially dangerous activities that require increased concentration of attention and speed of psychomotor reactions.
