Pharmacological action
Propofol has a general anesthetic effect.
Propofol is a drug intended for general anesthesia, which has a short-term and rapid effect (the effect of the drug develops over 30-60 seconds) and does not show a primary stimulating effect.
Lowers arterial, intraocular and intracranial pressure, total peripheral vascular resistance, minute breathing volume, and cerebral blood flow.
The effect of entering sleep is caused by a non-specific effect on the ion channels of neuronal membranes in the central nervous system. In the brain, the drug increases GABA-ergic processes. As a rule, recovery from anesthesia is not accompanied by nausea, headache and vomiting.
In pharmaceutical doses, it does not inhibit the hormonal synthesis of the adrenal cortex.

How to take, course of use and dosage
For introductory anesthesia, instructions for use Propofol involves intravenous use of the drug to adults, depending on weight.
At an average body weight, the recommended dose is 40 mg every 10 seconds, until the onset of clinical symptoms of anesthesia. Most often, the total dose of anesthesia remains at the level of 2-2.5 mg/kg.
In children after 8 years of age, the dose of the drug solution is calculated according to body weight (on average,2.5 mg / kg).
Intravenous use of the drug is carried out slowly, up to the occurrence of clinical symptoms of anesthesia. The dose for patients under 8 years of age may be higher.
People over 3 years of age who are qualified for ASA grades 3 and 4 are recommended to prescribe Propofol in smaller dosages.
In order to maintain the state of anesthesia, an infusion is administered at a dose for adults of 4-12 mg / kg per hour. Children’s dosage on average is 9-15 mg / kg per hour. It is also possible to repeat bolus use of the drug in the doses required to maintain adequate anesthesia.
To ensure the sedative effect of propofol during mechanical ventilation, adults are prescribed 300 mcg/kg per hour.
Special instructions
Application of the drug Propofol, as a rule, requires additional use of analgesics.
It should be used with caution in patients suffering from epilepsy (due to the risk of seizures), diseases of the cardiovascular, respiratory systems, kidney and liver diseases.
