Pharmacological action
Pharmacodynamics
Allochol refers to drugs that increase the formation of bile. The mechanism of action is due to reflexes of the intestinal mucosa, as well as the effect of Allochol on the secretory function of the liver. Allochol increases the amount of bile secreted, enhances the secretory and motor functions of the gastrointestinal tract, reduces the processes of putrefaction and fermentation in the intestines. Allochol can serve as a means of replacement therapy for endogenous bile acid deficiency.
Pharmacokinetics
Allochol is well absorbed in the gastrointestinal tract. Cholic and chenodeoxycholic acids contained in Allochol undergo 7α-dehydroxylation in the intestine. Chenodeoxycholic acid, in addition, undergoes metabolism in the liver: it conjugates with amino acids, is secreted into the bile, from where it is again released into the intestines, then partially reabsorbed; the rest is excreted in feces.
Indications
It is used for chronic hepatitis, cholangitis, cholecystitis and habitual constipation associated with intestinal atony.
Contraindications
- individual intolerance,
- acute hepatitis,
- acute and subacute liver dystrophy,
- obstructive jaundice,
- children under 7 years of age (for this dosage form).
Side effects
When using the drug in some patients, diarrhea and allergic reactions are possible. In these cases, the drug is canceled.
Interaction
Preparations containing aluminum hydroxide, cholestyramine, and cholestipol reduce absorption (and reduce the effect) — simultaneous use is impractical.
How to take, course of use and dosage
Taken orally after a meal. Assign adults 1-2 tablets 3-4 times a day. The course of treatment is 3-4 weeks.
In case of exacerbation of diseases,1 tablet is prescribed 2-3 times a day (for 1-2 months).
Overdose
Possible diarrhea, nausea, heartburn, itching of the skin, increased levels of transaminases in the blood.
Special instructions
The course of treatment with Allochol can be repeated 2-3 times with a break of 3 months.
